چکیده

It has been demonstrated by studies in vitro and in vivo that two absorptive processes are involved in the intestinal absorption of amino acids. It is also apparent that the vitamins of the Bggroup play a role in the “active” process of cellular uptake of amino acids (2) and that deoxpyridoxine (3) and 2,4-dinitrophenol (3,4), have an inhibitory effect upon active absorptive processes. We are presenting evidence that pyridoxal phosphate alleviates deoxypyridoxineand 2,4-dinitrophenol-induced inhibition of methionine absorption, and that the inhibitory block is linked to the phosphorylation of the vitamin cofactor. These studies extend our investigations on methionine absorption (5) and lend further support to the hypothesis that the Be-vitamins are involved actively in the absorptive process in the steady state.

Inhibition of in Vivo Conversion of Methionine to Ethylene by l-Canaline and 2,4-Dinitrophenol.

l-Canaline, a potent inhibitor of pyridoxal phosphate-mediated reactions, markedly inhibited the conversion of methionine to ethylene and carbon dioxide by apple tissue. A 50% inhibition of methionine conversion into ethylene was obtained with 50 mum canaline and almost complete inhibition with 300 mum canaline. When 2,4-dinitrophenol, an oxidative phosphorylation uncoupler, was fed to apple ti...

Vitamin B6 influences glucocorticoid receptor-dependent gene expression.

We have examined the influence of intracellular vitamin B6 concentration on glucocorticoid receptor function in HeLa S3 cells transfected with a glucocorticoid-responsive chloramphenicol acetyltransferase (CAT) reporter plasmid. CAT activity is induced from this plasmid specifically by glucocorticoid hormones in a glucocorticoid receptor-dependent manner. The intracellular concentration of pyri...

Transport and metabolism of vitamin B6 in the yeast Saccharomyces carlsbergensis 4228.

Active transport of pyridoxine, pyridoxal, and pyridoxamine occurs in resting cells of Saccharomyces carlsbergensis 4228 and can lead to intracellular concentrations of free vitamin much higher than those supplied externally. The initial Km for pyridoxine uptake is 3.6 x 10(-7) M at 30 degrees and pH 4.5, which are optimum for growth. Transport is inhibited by many unphosphorylated vitamin anal...

Purification and characterization of vitamin B6-phosphate phosphatase from human erythrocytes.

Human erythrocytes rapidly convert vitamin B6 to pyridoxal-P and contain soluble phosphatase activity which dephosphorylates pyridoxal-P at a pH optimum of 6-6.5. This phosphatase was purified 51,000-fold with a yield of 39% by ammonium sulfate precipitation and chromatography on DEAE-Sepharose, Sephacryl S-200, hydroxylapatite, and reactive yellow 86-agarose. Sephacryl S-200 chromatography and...

Studies on the nutrition of marine flatfish. The pyridoxine requirement of turbot (Scophthalmus maximus).

1. Diets containing graded levels of pyridoxine hydrochloride (to supply 0.26--30 mg pyridoxine/kg) were given to seven duplicate groups of turbot (Scophthalmus maximus) for 12 weeks and their growth rate was measured during this period. 2. Good growth was obtained on all treatments except those groups given less than 1.0 mg pyridoxine/kg diet. These fish grew normally until weeks 8--10 but the...
